What is the hybrid 90 degree function in microstrip?

The standard hybrid have four ports:
1. input port
2. output -3dB 90 degree phase shift compared to port 1
3. output -3dB 180 degree phase shift compared to port 1
4. port isolated
They are use among other things in balanced amplifier configurations. To achieve good return loss and high stability.

so if hybrid 180 degree, is it only use 1 and 3? port 2 neglect it by not connect to anything?
or how to configure which is 90 degree and which is 180 degree hybrid?

The branch-line coupler (90 degree hybrid)
Need all the ports to be connected to function correctly.
If it is a 50 ohm system, then all ports must be matched to 50 ohm. Otherwise you will not have the equal power spilt to port 2 and 3 and perfect match at all ports at the same time.
A 180 degree coupler (Rat-race) is able to make a 0 and 180 degree power spilt with the signals out of phase.
