How to measure antenna gain using Agilent VNA?

Hi, I have some questions in performing antenna gain measurement.
I am using Agilent Network Analyzer.
I have two questions as follows:
1. If you are used to Agilent Network Analyzer, I would like to ask the whether I should use B or B/R or B/R - M to obtain the value for gain calculation. I am very confuse.
2. Is calibration needed for gain measurement?
For your information, I am using Absolute-Gain Measurement (2 identical antennas).
